ارزیابی یک روش کچرز اصلاح‌شده جهت استخراج دیازینون و ادیفن‏فوس از دو خاک‏ مختلف شالیزارهای برنج با استفاده از HPLC-UV

چکیده

در این پژوهش، کارایی استخراج دیازینون و ادیفن‌فوس از دو خاک شالیزار با درصدهای ماده آلی متفاوت توسط یک روش کچرز اصلاح‌شده بررسی شد. روش‌ها شامل استخراج 10 گرم خاک با استونیتریل بدون افزودن و همراه با افزودن آب به نمونه‌ها، استخراج 5 گرم خاک با استونیتریل همراه با افزودن آب به نمونه‌ها و استخراج 5 گرم خاک با اتیل استات همراه با افزودن آب به نمونه‌ها بودند. خالص‌سازی به کمک روش استخراج فاز جامد پخش شونده انجام شد. اندازه‌گیری کمی آفت‌کش‌ها با استفاده از کروماتوگرافی مایع و آشکارساز فرابنفش/مرئی صورت گرفت. بر اساس نتایج، درصد ماده آلی خاک تأثیر معنی‌داری بر کارایی استخراج دو آفت‌کش داشت. عواملی همچون کاهش حجم نمونه و افزودن آب به آن پیش از استخراج و استفاده از اتیل ‏استات به‌عنوان حلال تأثیر معنی‌داری در افزایش کارایی استخراج داشتند. بهترین درصدهای بازیابی (70 تا 110 درصد) و انحراف معیار نسبی (کمتر از 20 درصد) با استفاده از استخراج 5 گرم خاک توسط اتیل استات به همراه افزودن آب به نمونه‌ها به ‏دست آمد. نتایج کالیبراسیون حاکی از خطی بودن این روش در محدوده غلظت‌های 5/0 تا 10 میکروگرم بر گرم بود. حساسیت مناسب روش برای دو آفت‌کش در دو خاک با توجه به مقادیر حد تشخیص و کمی شدن تخمینی (07/0 تا 54/0 میکروگرم بر گرم) تأیید شد.

In this research, the efficiency of diazinon and edifenphos extraction from two paddy soils with different organic contents was investigated using a modified QuEChERS extraction method. The methods included extraction of 10 g soil using acetonitrile with or without the addition of water to the samples, extraction of 5 g soil using acetonitrile with the addition of water to the samples, and extraction of 5 g soil using ethylacetate with the addition of water to the samples. The clean-up was carried out using dispersive solid phase extraction (dSPE). The pesticides quantification was performed using a liquid chromatography system equipped with a UV/VIS detector. According to the results, soil organic matter contents affected the recoveries of the two pesticides significantly. Factors such as reducing the sample size to 5 g, hydrating of the samples prior to extraction, and using ethylacetate as the solvent increased the extraction efficiencies of the two pesticides significantly. Best recoveries (70-100%) and relative standards deviations (RSDs) (˂20%) were achieved using 5 g soil as the sample with the addition of water and extraction using ethylacetate. The method had a good linearity within the range of 0.5-10 µg g-1. The sensitivity of the method was confirmed by the estimated method detection and quantification limits between 0.07-0.54 µg g-1.
