تأثیر بازدارندگی پایری‌پروکسی‌فن بر برخی از شاخص‌های فیزیولوژیک کفشدوزک Adalia decempunctata L. (Col.: Coccinellidae)

چکیده

کفشدوزک Adalia decempunctata L. مشهور به کفشدوزک ده­نقطه‌ای به­عنوان شکارگری فعال هم در مرحله لاروی و هم حشره کامل محسوب می­شوند. این حشره از روی انواع درختان میوه هسته­دار، دانه­دار، درختان جنگلی پهن­برگ و سوزنی­برگ آلوده به آفاتی نظیر شته، شپشک و پسیل از مناطق مختلف جهان گزارش شده است. پرورش کلنی این شکارگر در شرایط آزمایشگاهی (دمای ۲±۲۵ درجه سلسیوس، رطوبت نسبی 5±۷۵ درصد و دوره نوری ۱۶ ساعت روشنایی و ۸ ساعت تاریکی) انجام شد. در این پژوهش، آزمایش زیست­سنجی با یکی از تنظیم­کننده­ها­ی رشد حشرات به­نام پایری­پروکسی­فن به­روش موضعی مورد بررسی قرار گرفت. غلظت­های IC30, IC10 IC50, و IC90به ترتیب 6/18، 1/33، 3/49 و 6/130 میکروگرم بر میلی­لیتر تخمین زده شد. هم­چنین برخی از مهم­ترین شاخص­های بیوشیمیایی در لاروهای سن چهارم تیمار شده با غلظت­های بازدارنده­ی پایری­پروکسی­فن مورد ارزیابی قرار گرفت. نتایج نشان داد که میزان لیپید و پروتئین نسبت به شاهد به­طور نسبی افزایش ولی میزان گلیکوژن در لاروهای تیمار شده کاهش معنی­داری یافت. فعالیت آنزیم­های سم­زدا (گلوتاتیون اس_ترانسفراز و استرازهای عمومی) در لاروهای تیمار شده به­طور معنی­داری نسبت به شاهد افزایش یافت. آنزیم پراکسیداز نیز فعالیت افزایشی معنی‌داری را نسبت به شاهد نشان داد. با توجه به اینکه پایری­پروکسی­فن دارای ماهیت هورمون جوانی است، بنابراین مراحل نابالغ شکارگر مذکور تحت تأثیر قرار می­گیرند؛ در حالی­که حشرات کامل به دلیل گذار از این مرحله به هورمون رشد وابسته نبوده و کم­تر تحت تأثیر قرار می­گیرند. بنابراین بررسی­های تکمیلی در خصوص استفاده هم­زمان از پایری­پروکسی­فن و شکارگر فوق در شرایط صحرایی لازم می­باشد.

چکیده [English]

The lady beetle Adalia decempunctata L. (Col.: Coccinellidae) is known as the ten-spotted ladybeetle, whose larvae and adults are active hunters. These insects have been reported from several stone-fruit trees, seed-fruit trees, broad leaved forest trees and coniferous forest trees infested with pests like aphids, mealybugs and psyllid from several countries on the world. The culture of this predator was maintained in laboratory condition (25 ± 2 ºC, 75 ± 5% RH and 16:8h L:D). In this research, the bioassays of pyriproxyfen was performed through topical application method. The concentration of IC10, IC30, IC50 and IC90 were estimated 18.6, 33.1, 49.3 and 130.6 µg/ml, respectively. Also, in order to have a better understanding, some of the most important biochemical parameters were evaluated in the 4th instar larvae treated with sublethal concentrations of pyriproxyfen. The results showed that lipid and protein amount were relatively increased compared to control, while the glycogen amount was reduced in treated larvae. The activity of detoxifying enzymes (Glutathione S-transferase and general esterases) in treated larvae were significantly increased compared to control. The activity of peroxidase was also increase significantly compared to controls. As the pyriproxyfen is a juvenile hormone analogue, therefore the growing stages of this predator is affected. While adults that have surpassed this stage are no longer dependent to growth hormones and are less affected. Hence, supplementary evaluation of the use of pyriproxyfen and this predator at the same time under field conditions is needed.
